Human YAP2 Xpress Clones express YAP2, a transcriptional co-activator and key component of the Hippo pathway, which regulates cell proliferation, apoptosis, cancer, and organ size control. YAP2 interacts with Lats2 kinase, and when phosphorylated, it translocates to the cytoplasm, reducing its transcriptional activity. The YAP2-5SA mutant, where five serine residues (including S127) are mutated to alanine, cannot be phosphorylated, resulting in increased nuclear localization and higher basal activity. The YAP2-5SA-ΔC mutant lacks the C-terminal transcription activation domain, making it resistant to Hippo pathway-induced cytoplasmic translocation, and unable to activate gene expression. This form can act as a dominant-negative and restore contact inhibition in human cancer cells.
